09-P022 An ex vivo model of placental development
نویسندگان
چکیده
The placenta facilitates nutrient and gas exchanges between the mother and growing fetus. It does this through highly branched embryonic blood vessels called villi. In the mouse, villi start to form when the allantois attaches to the chorion. This attachment is mediated through a thin layer of mesothelium on the chorion, which becomes indistinguishable from the allantoic cells after attachment. Abnormalities in placental development underlie human pathologies such as pre-eclampsia, miscarriages, and intra-uterine growth-retardation. The genetic and structural features of the human placenta are conserved in mice, and the murine placenta is well characterized invivo. However, there are no systems to study and manipulate the placenta exvivo. We hypothesized that co-cultures of pre-attachment chorions and pre-attachment allantoises will mimic early invivo events in placental development, under the correct culture conditions. Chorions and allantoises of wild type mouse embryos were dissected at somite stage 3–6, and were combined and cultured for 12, 24, and 36 h exvivo. We found that under ourculture conditions, the allantois attachedto the chorion, and the mesothelium was indistinguishable from the allantois. We will present our morphological and molecular characterization of this exvivo model. This model will further our knowledge of early placental development as it allows for invitro manipulation of the allantois and chorions prior to culture.

Determination of the transport rate of xenobiotics and nanomaterials across the placenta using the ex vivo human placental perfusion model.
Decades ago the human placenta was thought to be an impenetrable barrier between mother and unborn child. However, the discovery of thalidomide-induced birth defects and many later studies afterwards proved the opposite. Today several harmful xenobiotics like nicotine, heroin, methadone or drugs as well as environmental pollutants were described to overcome this barrier. With the growing use of...
متن کاملEx Vivo Human Placental Transfer of Anti-Human Immunodeficiency Virus Compounds
OBJECTIVE The transfer of anti-human immunodeficiency virus (HIV) drugs has been studied in the ex vivo human placental model. There is a paucity of information on the placental transfer of these drugs because of ethical considerations and the expense involved in the use of the non-human primate model. METHODS The standardized ex vivo human placental model was used in these studies and the cl...

Background: Osteoarthritis is one of the most commonly diagnosed malignant diseases in the world and was previously recognized as a degenerative joint disease characterized by cartilage deformity and the formation of new bones at the edges of the joint. But this disease is not just a degenerative disease but also a biomechanical, biochemical, and cellular phenomenon. Common treatments include s...
متن کاملAntifungal potential of tolnaftate against Candida albicans in the treatment of onychomycosis: development of nail lacquer and ex vivo characterization
Onychomycosis constitutes the most common fungal infection of nail affecting finger and toe nails as well. Antifungals found to be effective in the treatment of onychomycosis. However, transport of oral antifungal agents exhibits more toxicity and requires longer treatment period. Medicated nail lacquers proved to cause lesser toxicity and required shorter treatment period. It provides not only...
